BioNuesis contains natural human extracellular matrix proteins including laminin, collagen IV and fibronectin. Full characterization of BioNuesis has not yet been completed.

Applications

BioNuesis is intended for the adherence and expansion of cells in culture, including stem and progenitor cells, such as human ES cells. Many other cell types which require the ECM proteins laminin, fibronectin or collagen type IV can also be cultured with BioNuesis.

Production of Human Extracellular Matrix for Stem Cell Culture Applications

Primary human dermal fibroblasts in scalable fermentation-style bioreactors grown in defined, animal-component free medium, have been used in the production of BioNuesis. In addition to the typical fibrillar collagens I and III, many ECM proteins with a known role in tissue development and regeneration are also produced, including the basement membrane components laminin and collagen type IV, along with fibronectin, tenascin and collagen type V, a fibrillar collagen only found at high abundance during fetal development. This hypoxic culture system also enhances the secretion of a wide variety of regeneration-associated growth factors and key developmental regulators of stem cell pluripotency and differentiation, such as the wnt gene family. This product technology offers a commercially viable source of human ECM for a wide variety of in vitro applications, including pluripotent embryonic stem cell culture, and potentially even directed differentiation of stem cells towards a particular lineage.
